Survival rate of hyperthyroid and hypothyroid rats with transplantable ascitic Zajdela’s hepatoma

Abstract

Introduction.The nongenomic actions of thyroid hormone begin at receptors in the plasma membrane, mitochondria or cytoplasm. Nongenomic actions initiated at the plasma membrane by thyroid hormone via integrinαvβ3can induce gene expression that affects angiogenesis and cell proliferation, therefore, both nongenomic and genomic effects can overlap in the nucleus. Scientific data demonstrate that excess concentrations of thyroid hormone are factors accelerating the natural history of cancer development. On the contrary, hypothyroidism for lymphoproliferative diseases and hepatocellular cancer is a risk factor for cancer progression.Aim.In the present study, the effect of drug-induced hypo- and hyperthyroidism was evaluated on the survival rate of rats with transplantable Seidel’s ascites hepatoma.Materials and methods.This study examined the effect of hyper- and hypothyroidism on the survival of Seidel’s ascites hepatoma bearing rats. Hepatoma cells were implanted intraperitoneally in male rats (n=48). Hypo- and hyperthyroidism was induced by oral administration of propylthiouracil (PTU) and L-thyroxine (Т4) 1.0 and 5.0 mg/kg body weight daily, respectively. Comparison group (n=12) were treated with cisplatin intraperitoneally one-time of 20,0 mg/kg body weight.Results.Survival analysis showed that cisplatin administration increased the life expectancy of laboratory animals by 32.1%, while hypo- and hyperthyroidism decreased the life expectancy by 4.8 and 10.7%, respectively. The median survival time of the euthyroid group was 8.0 (7.41–8.59) days, median survival time of the comparison group was 9.0 (8.78–10.2) days. Median survival time of the hypo- and hyperthyroid groups were 6.0 (5.75–6.92) and 6.0 (5.33–6.67) days, respectively (p=0.304).Conclusions.PTU-induced hypothyroidism and T4-induced hyperthyroidism leads to a decrease in the survival rate of laboratory animals with transplantable Seidel’s ascites hepatoma compared to the euthyroid group.
