Аннотация
Кожа — крупнейший орган, составляющий до 20% массы тела, ее барьерная функция обеспечивается постоянным обновлением. Стволовые клетки кожи поддерживают гомеостаз, регенерацию волос и заживление ран. В коже выделяют несколько типов стволовых клеток: межфолликулярного эпидермиса, волосяного фолликула (в луковице, корне, воронке), сальных и потовых желез. Формирование пула стволовых клеток начинается в эмбриогенезе, закладывая основу для регенерации во взрослом организме. Обновление эпидермиса долго объясняли стохастической (случайное самообновление) и иерархической (редкие деления стволовых клеток с образованием полустволовых клеток) моделями. Современные данные секвенирования единичных клеток привели к линейно-иерархической модели с множественными состояниями предшественников. Волосяной фолликул циклически проходит анаген (рост), катаген (дегенерацию) и телоген (покой). Предложена телескопическая модель его развития. У человека восстановление волос имеет отличия от мышиной, включая признаки эпителиально-мезенхимального перехода. В нише фолликула присутствуют стволовые клетки меланоцитов, обеспечивающие пигментацию; их пул пополняется клетками, возвращающимися из транзиторно-амплифицирующего состояния. Стареющие меланоциты через остеопонтин стимулируют активность соседних стволовых клеток волосяного фолликула. Стволовые клетки потовых желез (преимущественно эккринных) изучены слабее. В эмбриогенезе выявлены мультипотентные стволовые клетки, дающие начало миоэпителиальным и секреторным клеткам. У взрослых протоки обновляются быстро (как эпидермис), а концевой отдел — значительно медленнее. Нарушение баланса между покоем, самообновлением и дифференцировкой эпидермальных стволовых клеток ведет к истощению пула, плохому заживлению ран и кожным патологиям. Модуляция молекулярных путей для усиления регенеративного потенциала стволовых клеток открывает перспективы в лечении ран, возрастных изменений, восстановлении волос и косметологии.
